In March 1983, Earth was subjected to cosmic rays that gave sociopaths superpowers, resulting in a rise of super-villains known as Miscreants. With no one able to stop the Miscreants, normal people are usually left living in fear of being attacked by them. After a Miscreant kills her geneticist parents on their way home from work, Emily Stanton (Octavia Spencer) becomes determined to find a way to stop Miscreants. By 1985, Emily has sacrificed much of her social life in favor of researching possible methods of fighting back against Miscreants. While this results in her being bullied, Lydia Berman (Melissa McCarthy) stands up for her and becomes her best and only friend. Lydia is dumb as a rock, but loyal to Emily. Lydia supports Emily's dream of giving superpowers to normal people, though she tries to make sure that Emily doesn't overwork herself. When Lydia convinces Emily to take a half-hour nap from studying in 1993, she accidentally causes Emily to oversleep and wake up late for an AP exam, straining their relationship and causing them to drift apart. In 2024, Emily and Lydia have gone their separate ways, with Emily becoming a successful scientist and researcher for her own company and Lydia becoming a longshoreman. Lydia eats her meals at a cafe run by Frank (Kevin Dunn). Frank and Lydia worry that the miscreants are winning the war and wonder what Emily has been up to. Lydia tries to reconnect with Emily when their high school reunion comes around and invites her to come. When she fails to show up on the night of the reunion, Lydia concludes that Emily is still uncomfortable attending parties by herself and goes to pick her up. Emily tells her that while she would have liked to go to the reunion, she had forgotten when the reunion was and had a project she had to work on that night, which Emily wants to show to Lydia. Unfortunately, Lydia accidentally injects herself with a serum Emily had been working on. Lydia spilled her beer on Emily and when she went out to change clothes, Lydia activated the auto inject mechanism in Emily's lab that pushed the serum in her. The serum Emily injected was for super strength. Allie (Melissa Leo) is ex-CIA and helped Emily set up the whole lab. Having been injected with the serum, Lydia learns from Emily that the serum was designed to give a normal person superhuman strength and that she would have to undergo special training and treatment so the serum doesn't kill her (once the process is started, it has to be completed else the body will explode in a couple of days). Emily also joins Lydia in the treatment, although a less painful one since Emily took the serum in a pill form to take the other superpower concocted, invisibility. During the thirty-three-day treatment and training, Lydia and Emily grow closer. Lydia can only eat raw meat. Lydia discovers that in the years since their high school graduation, Emily had a daughter, Tracy (Taylor Mosby), with one of the researchers, who could not handle the responsibility and left them. Lydia plays online video games and bonds with Tracy, but Emily tells Tracy not to goof off. Once they finish their training and treatment, Emily (Bingo) and Lydia (The Hammer) foil a liquor store robbery run by a Miscreant with crab arms known as the Crab (Jason Bateman), who falls in love with an equally smitten Lydia, much to Emily's concern. The girls use a purple Lambo as their ride, a car that they are too fat to get into. Emily and Lydia, known as the superhero team Thunder Force, are praised for their heroics. This brings Thunder Force to the attention of mayoral candidate William "The King" Stevens (Bobby Cannavale), whose campaign is built on the idea that only he can end the Miscreants' crimes. The King has attacked by a Miscreant called Laser, many times during the campaign, bolstering his credentials. Secretly the King has Crab and Laser on his payroll. The King is also a miscreant himself. With the help of Laser (Pom Klementieff), a Miscreant who can generate and control whip-like energy beams, the King tries to get Thunder Force to work for him, leaving Chicago at the mercy of the Miscreants unless he wins the mayoral election. Thunder Force continues to fight crime with their superpowers and support the rival mayoral candidate Rachel Gonzales (Melissa Ponzio), thereby causing the King to lose the election. The King then sends Laser to attack Thunder Force when they're at a diner. When she tries to get away, Lydia throws a bus at her despite Emily's protests. Though nobody was hurt, Emily decides that Lydia's impulsiveness is more problematic than beneficial, once again straining their friendship. In an effort to make amends, Lydia goes on a date with the Crab to get some useful information. Along with how the Crab is a misunderstood person who had to turn to a life of crime, Lydia learns that the King is planning on blowing up everyone who did not vote for him in the election, along with the new mayor, at a party he's hosting under the guise of a celebration of the new mayor. She tells Emily of what she learned and makes amends with her. After fighting off Laser one last time (Allie had secretly made a deal with the King and told them about the Thunder Force's plan to infiltrate the building and steal the bomb. King sent Laser to fight them off), Thunder Force go to stop the King from bombing the building. The King figures out that the Crab told Lydia about his plans, but doesn't fire him. When they manage to find the bomb, the King decides to fight Thunder Force himself, revealing himself to be a Miscreant with superhuman strength, though significantly stronger than Lydia. Before the King can kill Lydia, the crab double crosses him and gets his claws broken off. Tracy also joins the fight, having injected herself with her mother's serum, giving her the ability to run at superhuman speed. Though they manage to take the King down, Thunder Force realizes that the bomb will go off before they can disarm it. With no guarantee that the bomb would be stable enough to not go off while Tracy carries the bomb to somewhere safe, Lydia decides to sacrifice herself, knowing that she can at least reduce the impact of the explosion. She jumps out the building with the bomb and dives into the Chicago River, seemingly dying in the explosion. However, the paramedics manage to find her body and resuscitate her. Thunder Force, now with a stronger friendship between Emily and Lydia, are offered by the mayor the assistance of the city's resources, which they accept.

Wendy (Dakota Fanning) - a young woman with autism and a fixation on Star Trek - lives a routine life in a San Francisco group home where she is monitored by the house manager, Scottie (Toni Collette). She spends her time writing a 450-page Star Trek script to enter in Paramount Pictures' writing competition in hopes of winning the $100,000 prize. Wendy is visited by her sister Audrey (Alice Eve), who shows her pictures of her infant daughter, Ruby, and reveals that she is selling their childhood home. Wendy asks Audrey to take her home, arguing that she will be able to buy back the house and help care for Ruby once she wins the screenwriting contest. Audrey refuses, informing Wendy she is not capable of caring for a baby. Unfortunately, Wendy has a meltdown and she leaves the group home weeping. After Wendy misses the mail-in date to send her script to Paramount Pictures, she decides to deliver the script personally. She leaves the group home early the next morning and is followed by the group home's small dog, Pete. Wendy and Pete board a bus to Los Angeles, but are kicked off and left by the side of the road after Pete urinates on the bus. Wendy then wanders into a shanty town, where she is robbed of most of her money. She then wanders to a nearby shop and is nearly swindled out of the last of her money by an employee. However, an elderly woman named Rose (Marla Gibbs) intervenes to stop him from exploiting Wendy. She sympathizes with her as her grandson also has autism, and lets Wendy accompany her on the senior citizens' bus. However, the driver falls asleep at the wheel, crashing the bus without warning. Following the bus crash, Wendy wakes up in a hospital in Bakersfield. Still determined to deliver her script as planned, she leaves Pete at the hospital and escapes. During her escape, Wendy loses part of her script by chance. She gathers used paper and begins rewriting the script. At the same time, Audrey and Scottie have realized Wendy has gone missing. They begin to search for her and request a missing persons report. After correctly deducing that Wendy left for Los Angeles to personally deliver her script, Scottie and her son Sam (River Alexander) leave on the road. The Bakersfield hospital notifies Scottie and Audrey of Wendy's whereabouts; police continue the search from there. Scottie and Sam find the missing script pages while scouring the hospital. Wendy then attempts to buy a bus ticket to Los Angeles, but is unable to do so as she has no money. She ultimately stows away on the next bus there, hiding inside the baggage compartment. Upon her arrival to Los Angeles, Wendy wanders around until two police officers recognize her from the missing persons report. Fortunately, Officer Frank (Patton Oswalt) eventually gains Wendy's trust by speaking to her in Klingon. The police officers bring Wendy to the police station, where she is reunited with Audrey and Scottie. They drive Wendy to Paramount Pictures so she can deliver her script there as planned. Despite being stonewalled by a bad-mannered mailroom worker, Wendy is able to sneak past security and submit the script to the turn-in box. Satisfied that she has completed her mission, Wendy informs her sister she did this to prove that she was more capable than Audrey believed. Wendy returns to the group home where she later receives a letter from Paramount Pictures informing her that her script was not chosen, but encouraging her to continue writing. Despite the rejection, she is satisfied with everything she has accomplished. Wendy visits Audrey at their childhood home and holds her niece in her arms for the first time, while her sister dearly embraces her.

DArthur Denning (Hopkins) is the owner of a major pharmaceutical firm. He is very controlling of his girlfriend Emily Hynes (Åkerman), and she is physically intimidated by his security guards. She is abducted, and Denning is sent photos of her bruised face. He is ordered to go to an art gallery with the ransom. When a man approaches him, Denning attacks him, only to find out later that he was the gallery owner. Before her abduction, Hynes reaches out to her ex-boyfriend Ben Cahill (Duhamel). Over drinks, she hints at Denning's abuse and some dirt she has on him. Back at her apartment, she shows Cahill files that she has stolen from Denning's laptop that reveal his criminal behavior during drug trials. She attempts to seduce Cahill. Later, she convinces a stranger to beat her up for the photo that she sends to Denning. Cahill uses the information from Hynes to pitch a class action lawsuit to his boss Charles Abrams (Pacino) which he then files as a class action suit against Denning. Later that evening, Cahill is stunned to find Hynes dead in her apartment. With a bottle of pills in her hand, and a lethal cocktail of drugs in her system, her death is suspicious but possibly just a suicide. Cahill decides to leave the scene without reporting it to the police and tries to cover his tracks secretly telling only his wife about it. At a preliminary deposition to the lawsuit, Denning suddenly settles the suit for $400 million, provided that the stolen documents are returned to him. A short time later, Hynes body turns up in Cahill's home. Convinced he is being framed, he then confronts Denning who is amused at Cahill's passion, but assures him that he had nothing to do with it. A hitman kidnaps Cahill and his wife Charlotte (Eve). Cahill manages to overpower and kill him. When he inspects the hitman's files he realizes that the villain of the piece is actually his boss. Abrams has been colluding with Denning for years, deliberately losing all the cases his clients brought against the pharmaceutical magnate. The settlement was all just an elaborate way of protecting Denning from criminal liability. Cahill submits the hitman's files to the police who arrest Abrams. Before he can be taken into custody, Abrams grabs an officer's gun and commits suicide. As Cahill packs for a move to a new house, he notices that some of his wife's clothing smells like Hynes' perfume. He asks her what happened between the two of them. Charlotte confesses that she confronted Hynes about her involvement with her husband. They had an argument in which Charlotte hit Hynes, causing her to fall and hit her head. Though Charlotte is a nurse, she did not help her and staged the scene for the police.

A struggling motel owner and her daughter are taken hostage by a nearly blind career criminal to be his eyes as he attempts to retrieve his cash package from a crooked cop.

In 19th Century Baltimore, a killer terrorises the city by orchestrating a series of grisly murders which seem to mirror the killings from the dark tales of Edgar Allen Poe. When the police find themselves unable to stop this spree of murders, they enlist the help of Poe himself to assist them in nabbing the killer. The stakes are raised when Poe`s to be wife, Emily Hamilton is abducted by the killer and her life is put in jeopardy unless Poe writes his tales of terror again. As the police and Poe race towards trying to unmask the killer, time is running out for Emily.

A 50-year-old salaryman named Kenichi Shibuya has been divorced for five years and has an awkward relationship with his daughter Mika. The day before Mika's 15th birthday, he's killed by a bolt of lightning on the side of the road along with one of his superiors from the office, Ayase. In the afterlife, they're told they can be reincarnated if they drink a "legendary soup", but they'll lose all memories from their former life in the process. Refusing to accept the idea of forgetting his daughter, Shibuya instead tries to unravel the secrets of the soup by wandering around the afterlife with Ayase as he attempts to continue his fatherly role from the other side.

After immigrant Mireya Sanchez is deported, officer Max Brogan takes care of her little son, and brings him to the boy's grandparents in Mexico. Later the woman is found dead near the border. Brogan returns to the grandparents to tell them the bad news. Cole Frankel, a corrupt immigration officer, makes a deal with Australian immigrant Claire Shepard: he can have unlimited sex with her for two months, then she gets a green card. In the course of the two months he separates from his wife and wants to make the relationship with Shepard one of love, but she declines. He exempts her from completing the two months and arranges the green card. However, authorities find out and arrest him and Shepard is deported. His wife Denise Frankel adopts a little girl from Nigeria, who has already been in the detention center several years. Brogan has an Iranian colleague, Hamid Baraheri. Hamid's family disapproves of his sister having sex with a married man. Encouraged by his father his brother plans to beat them up, but he ends up killing them, and is arrested. South Korean teenager Yong Kim, who is about to be naturalized, reluctantly participates in a robbery with four others. Hamid kills the four in a shootout but lets him go. He lies to the authorities that there were only four robbers. Gavin Kossef, a young British immigrant, pretends to be a religious Jew, to get a job at a Jewish school, which allows him to stay in the U.S. In a test where he has to demonstrate his familiarity with the Jewish religion he does not perform properly, but a rabbi asked to assess it approves it, so that the immigrant passes the test. After the test, in private, the rabbi requires from the immigrant to take lessons from him to eliminate the deficiencies in his knowledge. Taslima Jahangir, a 15-year-old girl from Bangladesh, presents a paper at school promoting that people should try to understand the 9/11 hijackers. The school principal reports this to authorities. She is not charged for this, but it turns out that her parents and she are illegally in the U.S. One parent of choice can stay with the girl's two younger siblings, who are U.S. citizens because they were born in the U.S., the girl has to leave with the other parent to Bangladesh, even though she has lived there only until she was three. She leaves with her mother, and cannot even say goodbye to her father.
