Saul says he may know Dale's next customer, to whom Dale is serving some court papers, Ted Jones (Gary Cole). Saul buys his supply from Red, who buys it from Ted. At Ted's house, Dale is waiting outside to finish his joint when a police car pulls up. Dale witnesses Ted and a police officer, Carol Brazier (Rosie Perez), shoot and kill a rival Korean gangster. While fleeing, Dale leaves behind his roach (joint) containing Pineapple Express, the rare strain of marijuana he just purchased from Saul. Ted identifies the strain and sends his henchmen Budlofsky (Kevin Corrigan) and Matheson (Craig Robinson) to Red (Danny McBride), a dealer who tells them about Saul. Dale rushes back to Saul & at his apartment, Dale learns Ted is a drug lord and could trace the roach (as only Red and Saul have the Pineapple Express strain). Dale and Saul decide they must flee the city. They attempt to hide in the woods where Dale's car battery dies. Dale and Saul visit Red, who was already working with Budlofsky and had ratted Saul out. Red reveals that Ted knows who they are and intends to kill them. Dale and Saul escape, as Budlofsky comes in. Budlofsky and Matheson torture Red for information and both shoot him in the abdomen. Ted orders Budlovsky to kill Red. Meanwhile the Korean gang members are outside Ted's house and listening to all his conversations. Believing Dale's high-school girlfriend Angie to be in danger, he and Saul travel to her home. Dale shows up for dinner at his girlfriend's Angie (Amber Heard) parents' house and obviously it goes bad as Dale is bleeding and he stinks. Angie's dad pulls out his shotgun and starts shooting at Dale & Saul, but Dale manages to convince him to calm down. As Budlofsky reaches Angie's house (tracking Dale's car), Dale and Saul leave. Angie's family goes to a hotel. Meanwhile Ted and Carol talk to the Korean mob boss Cheung (Dana Lee) and tell him that his assassin is dead. Ted promises retaliation if Cheung does not withdraw Dale. Ted thinks Dale is another assassin sent by Cheung. Cheng decides to attack Ted. Dale and Saul decide they must leave the city. They sell Pineapple Express to raise bus fare, but Dale is arrested by Police Liaison Officer Bobbra (Cleo King). Dale tells Bobbra that he witnessed Brazier and Ted murder a man. Bobbra has long been suspicious of Brazier's corruption and says she will investigate. However, Saul, thinking that it is Brazier driving, hijacks the police car. Brazier hears on police radio of Dale's arrest and pursues Dale and Saul in a high-speed chase, but they escape. After Dale and Saul argue, they call each other stupid and the main reason of why they both are in this mess. Dale and Saul split up, and Saul is kidnapped. Budlofsky was waiting at Saul's grandmother's retirement home, when Saul visited her. Saul is held in Ted's lair beneath the same military facility from 1937. Dale enlists Red's help to rescue Saul. Red had miraculously survived 2 gunshots to his body and arms himself and Dale and drives Dale to the lair where Saul has been kept. Red backs out at the last minute and Dale is captured by Budlofsky. While Dale and Saul are waiting to be killed, they reconcile and plan an escape. Korean mobsters attack the barn to avenge their fellow gangster's death, the murder that Dale witnessed. Dale and Saul free themselves but are caught by Matheson (Craig Robinson). Matheson is Budlofsky's assistant and has been working with this entire time. A brawl ensues between the attacking Asians and Ted's forces who retaliate. There are heavy casualties on both sides. Saul takes on Brazier, while Dale combats Ted. Both manage to come out on top, but Matheson has the gun on Saul. Red reappears and drives his car through the barn, killing Matheson and saving Saul, but is shot by Brazier. A mobster sets off a bomb killing Ted and setting fire to the barn. Red's car explodes and lands on Brazier, killing her. Dale carries Saul from the burning barn and Red, wounded but still alive, also escapes and reconciles with them. They talk about their adventure over breakfast at a diner before Saul's grandmother picks them up and takes them to the hospital.

Molly (Kate Hudson) and Carl (Matt Dillon) are preparing for their wedding day in Hawaii, until Carl's friend Neil (Seth Rogen) interrupts to say that Dupree (Owen Wilson) got lost. They drive off together to pick up Dupree, who appeared to have hitched a ride with a light plane after landing on the wrong island. A day before the wedding, Molly's father Thompson (Michael Douglas), who is also CEO of the company that Carl works for, makes a toast with humorous remarks at Carl's expense, foreshadowing a conflict between the two. Dupree laughed a bit too loudly at Thompson's jokes. Later at a Pre-celebration at a bar, Carl neglects Dupree to be with Molly (as she wanted to introduce Carl to her grandmother) while he is about to perform a drinking tradition called the "Flaming Tornado". Carl and Dupree later make up on the beach, as Dupree apologizes for laughing at Molly's father's jokes, and tells Carl that he has "Carlness." Dupree says that Carl has that optimistic bent of mind that will never let anything, or anyone get to him, and asks him never to lose that. Carl and Molly get married. When Carl returns to work, at Molly's father's Thompson Land Development, he is surprised to find that Mr. Thompson has promoted him to be in charge of a design he proposed, though it had been altered somewhat. Carl had proposed a small 40-unit community, but Thompson decides to super-size it to several hundred units. The project is named "Oaks at Mesa Vista", and yet the site selected by Thompson has no trees at all. Mr. Thompson makes absurd requests which proceed to get worse, starting with Thompson's drastic re-imagining of Carl's new architecture project, requesting that Carl take the Thompson name instead and that Carl get a vasectomy to prevent childbearing later on. Before returning home to celebrate his promotion with Molly, Carl stops by the bar, where he finds Neil and Dupree. After Neil leaves due to his wife's curfew, Dupree reveals that he was fired, as he apparently did not have clearance to attend the wedding (his boss claimed that he never got Dupree's email), subsequently falling behind in his rent and evicted. Losing his car also, as it was the company's. Dupree was planning to sleep in the back room at the bar, but some of the regulars complained that Dupree got to sleep at the bar, and they didn't, so the cot was removed. Carl takes Dupree home, asking Molly if he can stay for a little while until he gets back on his feet. Molly is polite, though clearly frustrated as he is disruptive and messy. We learn that Dupree was a copier salesman at his last job. The next morning, Molly and Carl come down from their room all dressed for work and find Dupree, still sleeping on the couch, naked. While Molly and Carl are at work, Dupree engages the neighborhood kids in a game of baseball and causes a brawl by encouraging the kids to play aggressively. Molly sets up Dupree with a woman at her work, a primary school, who is a Mormon librarian. Dupree agrees, though Molly is shocked to find them having sex when she comes home from dinner. Romantic candles that Dupree had set up burn down the front of the living room, and Dupree is kicked out. Meanwhile Carl is being continually stressed out from work, though he and Molly find time to go out for dinner. On the way back they find Dupree sitting on a bench in heavy rain with his belongings. Molly insists they take him back in. Carl makes it known that Dupree must behave this time. The next day, Dupree makes amends, refurbishing the living room, and doing Carl's thank-you letters, as well as making friends with kids from the block. Carl asks Dupree to go to career day at Molly's work as he got tied up at his work and that night Dupree cooks a large dinner for Molly, though Carl is late again, so Molly and Dupree start without him. When Carl finally shows up, he is a little jealous that they were having dinner together and have a fight. Carl kicks Dupree out, suspecting an affair, which shocks Dupree. The following night, Mr. Thompson is over for dinner. Dupree climbs the drain pipe and sneaks inside to get his bags back, and during dinner the guests inside hear Dupree fall off the roof. Dupree is found outside and is invited in for dinner. After Bob takes a liking to Dupree and asks him to go fishing with him, it enrages Carl because - even though he does not like fishing - it is Bob's way of approving someone. Carl's fury boils over as he then imagines Dupree hanging out with Bob and having sex with Molly, causing Carl to jump across the table and attempt to strangle Dupree. Thompson hits Carl over the head with a candlestick shortly after and Molly kicks Carl out of the house. After returning from the hospital with a neck brace, Dupree and Molly confront Bob about what he really thinks of Carl, and his evasive answers reveal his dislike of him. The next morning Dupree gets all the local kids to search for Carl. Dupree eventually finds Carl in the bar and convinces him to chase after Molly. Dupree helps Carl break into Mr. Thompson's office, as Dupree distracts Paco, the security guard, whilst Carl marches into his father-in-law's office and confronts him. The two finally reach an understanding and Thompson admits to his agenda of insulting Carl. Dupree and Carl return to the house, where Carl and Molly reunite, Carl apologizing, and agree to work it all out. Glad that he mended Carl's marriage, Dupree celebrates by leaping into the air with joy until he falls to the ground. Dupree uses his experiences with Carl and Molly to publish a successful self-help book titled 7 Different Kinds of Smoke: Living, Loving, and Finding your Inner "-ness" and becomes a motivational speaker, with Paco as his assistant. In a post-credits scene, Dupree's hero Lance Armstrong reads Dupree's book and repeats "Lanceness" with various inflections.

The movie begins as we meet Andy Stitzer (Steve Carell). He lives in a downstairs apartment and is unusually organized and tidy. He has a massive collection of action figures and video games. He also has a high tech video system and even a video chair for playing his games. This is the perfect 20 year old's apartment; unfortunately, he's 40. As he wakes up we watch as he struggles to pee with his "morning wood" and then makes a perfect breakfast omelet with fruit and juice. Again, everything is neat, organized and well planned out. He goes out and chit chats with the upstairs neighbor and his wife, making plans to come over to watch Survivor. He gets on his bike and rides to work. He works at a giant electronics store, think "Best Buy" or something similar. He works in the inventory control room, away from the public. He's actually a very nice guy but his co-workers think he's weird and possibly a serial killer. One day his coworkers discover they are one person short for a poker party they are planning later that night so they decide to ask Andy; he accepts their offer. Much to their surprise, Andy's great and beats them all. Everyone's sitting around swapping sex stories and when it comes to Andy he makes something up and it's obvious he's lying. They call him out and ultimately it is revealed that he is a virgin. Andy "rides" home in shame, completely embarrassed and ready to quit his job. He's up all night and just can't stop thinking about the humiliation that awaits him in the morning. The next morning he cautiously enters work and everything is fine at first and then one by one, different co-workers start making cracks about him. He runs out of the place in shame but one of his poker buddies stops him and eventually all his buddies get together and make it their personal goal to help him with his problem. For the next 45 minutes or so of the movie we see many different parties, clubs, and encounters he gets set up on, all pretty funny but none of them work. Among them by far the funniest is when they decide he needs to have his chest waxed. The torture he goes through is hilarious and gets even funnier when we see him walking home with a little blood on his shirt. At a bar one night they advise him on finding a drunk girl to take home. He does but since he doesn't have a car or license, the drunk girl drives, almost killing him and the evening ends with her throwing up on him. She offers to still have sex with him but he turns her down. One day at work he is forced to work out on the floor for a bit and meets Trish (Catherine Keener). Trish owns a small store across the street called "We Sell Your Stuff On eBay" The two hit it off and she gives him her phone number. He tells his buddies about her but they insist that he if he really loves her he can't have sex with her until after he loses his virginity. If his first time is with her it will be awful and she'll think he's a freak. Again they try to hook him up but nothing works. Finally after a date with Trish it looks like it's actually going to happen. They're on her bed making out when she goes into the bathroom to get ready. She tells him that there are condoms in the drawer but check the expiration date because they're pretty old. He starts reading instructions and such, not quite sure how to use one. He goes through a dozen or so just trying to get one on when suddenly Trish's daughter and boyfriend walk in. Her daughter is angry and she and her mom get into a huge argument. The boyfriend looks down on the what looks like a dozen used condoms laying on the bed and whispers to Andy, "teach me!" Andy and Trish decide to put off sex for awhile and just get to know each other. They decide to wait until their 20th date to have sex. They have many dates and Andy and Trish's daughter are actually becoming quite close. Trish helps him with his dream of opening his own electronics store by selling some of his collection at her eBay store. Things are going just great between the two of them. One day Trish goes over to Andy's apartment and finds a collection of porn that one of Andy's buddies gave him. Trish can't believe what kind of freak Andy is and rushes out. Andy chases after her and the chase ends with Andy crashing his bike into a billboard truck, landing right in front of her. As she is leaning over him he finally confesses to her that he is a virgin. The two have a passionate embrace. The next thing we know it's their wedding day. On the wedding night they finally make love. She tells him, congratulations, you are officially not a virgin. This encounter lasts 1 minute. She asks if he wants to do it again and he says sure... Two hours later we see them lying in bed with big grins on their faces. She asks him how it was and then the movie breaks into a strange video of him singing "Aquarius" with many of the cast of characters from the film in the background.

He works alongside his friends on the news team: fashion-oriented lead field reporter Brian Fantana (Paul Rudd), sportscaster Champion "Champ" Kind (David Koechner), and a "legally retarded" chief meteorologist Brick Tamland (Steve Carell). The team is notified by their boss, Ed Harken (Fred Willard), that their station has maintained its long-held status as the highest-rated news program in San Diego, leading them to throw a wild party. Ron sees an attractive blond woman and immediately tries to hit on her. After an awkward, failed pick-up attempt, the woman leaves. The next day, Ed informs the team that he has been forced by the network to hire Veronica Corningstone (Christina Applegate), a female news reporter from Asheville, North Carolina and the same woman Ron had tried to pick up the night before. The news team attempts to seduce Veronica using various inept, arrogant and sexist methods, all of which backfire. Ron ends up asking her out under the guise of helping out a new co-worker, which she accepts. During their date, Ron woos Veronica by playing jazz flute in his friend Tino's (Fred Armisen) club. Veronica goes against her policy of not dating co-workers and sleeps with Ron. The next day, despite agreeing with Veronica to keep the relationship discreet, Ron tells the entire news room that he and Veronica are in a sexual relationship (and later announces it on the air). The next day when Ron is on his way to the station, he throws a burrito out his car window, accidentally hitting a motorcyclist (Jack Black) in the head, causing him to crash. In a fit of rage, the biker retaliates by punting Ron's dog, Baxter, off a bridge. A distressed and incoherent Ron calls Brian from a pay phone and tells him what happened. Since Ron is now late, Brian frantically tells him to come to the station because Veronica is about to take his place. Despite Ron's efforts to arrive on time, Veronica goes on air. After Ron arrives, he has an argument with Veronica about the situation and they break up. The next day, Veronica is made co-anchor, much to the entire news team's disgust. The co-anchors soon become fierce rivals and argue with each other both on and off the air. One day while feeling down on themselves, the News team decides to buy new suits. However on the way to the suit shop Brick, who was leading the way, gets them lost ending up in a shady part of town. They are then confronted by Wes Mantooth (Vince Vaughn) and the evening news team. Tired of their rudeness and petty anger, Ron challenges them to a fight. All armed with crude weapons the two teams are about to fight when they are joined by Channel 2 news team with lead anchor Frank Vitchard (Luke Wilson), the public news team and their lead anchor (Tim Robbins), and the Spanish language news with lead anchor Arturo Mendez (Ben Stiller). A full on melee ensues between the five teams until they all flee when police sirens are heard. When they return to the studio, Ron gets in another heated argument with Veronica and they get in a physical fight after she insults his hair, increasing tensions even more. While in a restaurant celebrating Veronica's success, one of Veronica's co-workers tells her that Ron will read anything that is written on the teleprompter. Later, Veronica sneaks into the station and changes the words in Ron's teleprompter. The next day, instead of Ron saying his signature "You stay classy, San Diego!" closure, Ron ends the broadcast with, "Go f*** yourself, San Diego." Everyone in the studio, except Ron, is speechless. An angry mob gathers outside the studio and Ed is forced to fire Ron. Garth is especially hurt and sobs while labeling Ron "a poopmouth". Veronica sees that she has gone too far and attempts to apologize, but Ron angrily dismisses her while being led through the mob by security, yelling at her that she had "reduced him to rubble". Three months later, Ron is unemployed, friendless, hated by the city, and is a slovenly drunk. In this time, Veronica has become very famous, but is hated by her male coworkers for sabotaging Ron; Ed told Champ, Brick, and Brian not to talk to Ron or he will fire them if they do. When it is announced that Ling-Wong the panda is about to give birth, all San Diego news teams head to the zoo to cover the story. In an attempt to sabotage her, the public news anchor pushes Veronica into a Kodiak bear enclosure. When Ed cannot find Veronica, he calls the bar where Ron spends most of his time and reluctantly asks him to return. Ron then summons the rest of his team by blowing the "News Horn", despite the fact that they were all standing a foot away playing pool. Baxter, who has miraculously survived, hears this call and follows the voice to find Ron. Once at the zoo, Ron jumps into the bear pen to save Veronica, with everyone else at the zoo watching. The Channel 4 news team then jumps in to help Ron. Just as the leader of the bears is about to rip Ron and Veronica apart, Baxter shows up and convinces the bear to let them live by mentioning that he is a friend of the bear's cousin, Katow-jo, whom he met in the wild. After Ron and Veronica reconcile, it is shown that in years to come, Brian becomes the host of a Fox reality show named Intercourse Island, Brick is George W. Bush's top political adviser, Champ is a commentator for the NFL before he was fired for sexually harassing Terry Bradshaw, and Ron and Veronica are co-anchors for the CNN-esque World News Center, taking over after the narrator retires and it is shown to be some time later as Ron and Veronica are shown wearing 80's fashions.
